Summary of Position: 

Kronos Bio seeks a proteomics expert in protein-protein interaction mapping at the Scientist/Senior Scientist level to join its drug discovery team located in Cambridge, MA. As a member of an interdisciplinary team, this individual is expected to harness their background in proteomics and mass spectrometry to help map and drug protein complexes involved in transcription as a part of broader oncology drug discovery programs.

The applicant is expected to be a highly motivated individual with a strong technical background who works equally well driving projects forward independently and as part of a collaborative, multidisciplinary scientific team. The applicant will possess a robust skill set applicable to proteomics, drug discovery and target biology, with strong interpersonal skills, critical thinking and eagerness to strengthen their knowledge both within and beyond their area of expertise.

Essential Duties and Responsibilities:

• Design, develop, and execute Affinity Purification-Mass Spectrometry (AP-MS) experiment to profile protein-protein interaction network for TRN mapping efforts.
• Evaluate and optimize affinity purification, including lysate and pull-down conditions, using Label-Free Quantitation (LFQ) and TMT proteomics approach.
• Scale up and automate AP-MS for TRN PPI mapping of multiple targets in parallel to enable the confidence measurement of protein interaction using network topology strategy.
• Optimize and select appropriate proteomics strategy to capture transient and weak protein-protein interactions to drive drug discovery program.
• Implement the most advanced proteomics quantitation and data analysis workflow to increase the sensitivity and specificity of AP-MS data.
• Explore co-fractionation mass spectrometry to profile protein complex in cell lysate.

Qualifications:

• PhD in Analytical Chemistry, Biochemistry, or related discipline for the Scientist level or with 2+ years post-doctoral experience (industry or academia) for the Senior Scientist level.
• Possess a robust experimental skill set in proteomics with an emphasis on protein-protein interaction mapping.
• Proven ability to operate and trouble-shoot nano LC-MS instrument is required, ideally Thermo Orbitrap-based MS systems.
• Extensive experience in analyzing and interpreting LC-MS/MS data using custom, public, and commercially available software suites (Proteome Discover, MaxQuant, Perseus, MSstats, etc).
• Solid understanding of large-scale proteomics experiment study design and data analysis workflow.
• Experience in co-fractionation mass spectrometry is desirable.
• Experience with affinity purification and proteomics sample prep workflow automation using liquid handler is a plus.
• Ability to work independently and balance multiple tasks and/or projects.
• Significant experience developing and troubleshooting experimental workflows.
• Demonstrated ability to communicate scientific concepts and data informally and externally through presentation and publication.
