Generation Bio is innovating genetic medicines to provide durable, redosable treatments for people living with rare and prevalent diseases. The company’s non-viral platform incorporates a novel DNA construct called closed-ended DNA, or ceDNA; a unique cell-targeted lipid nanoparticle delivery system, or ctLNP; and a highly scalable capsid-free manufacturing process that uses its proprietary cell-free rapid enzymatic synthesis, or RES, to produce ceDNA. The platform is designed to enable multi-year durability from a single dose, to deliver large genetic payloads, including multiple genes, to specific tissues, and to allow titration and redosing to adjust or extend expression levels in each patient. RES has the potential to expand Generation Bio’s manufacturing scale to hundreds of millions of doses to support its mission to extend the reach of genetic medicine to more people, living with more diseases, around the world.

SUMMARY: 

Generation Bio seeks an energetic, versatile, and highly motivated Sr. Associate or Principal Associate Scientist to join our discovery research group. For this position, the successful candidate will support development of our cell targeted lipid nanoparticle (ctLNP) delivery platform. The ideal candidate has a background in antibody discovery as well as expertise in HTP in vitro, plate-based assay development. They will also have experience with basic protein expression and characterization. They will work with the broader discovery organization to identify as well as characterize structural and functional properties of antibodies and/or related molecules using diverse techniques. This role will be responsible for helping to interrogate novel cell targeting mechanisms and enabling cell specific uptake of our LNPs. Responsibilities will also include cross-functional research to enable early program development for indications in target cell types.


RESPONSIBILITIES:

  • Perform a range of biochemical techniques such as ELISA, MSD, Western blotting, and SDS-PAGE to characterize protein expression and function.
  • Perform a range of biophysical techniques such as surface plasmon resonance (SPR), biolayer Interferometry (BLI) and/ differential scanning calorimetry (DSC) to characterize protein-ligand interactions and protein stability.
  • Express and purify proteins in small scale for in vitro assays.
  • Carry out molecular and cell biology experiments, including but not limited to, DNA cloning, transient transfections, cell culture maintenance.
  • Assist to analyze and interpret data from biophysical assays to understand binding kinetics, thermodynamics, and binding affinities.
  • Assist to troubleshoot and optimize assays as needed to improve sensitivity, specificity, and reproducibility.
  • Maintain accurate and up-to-date laboratory records.

 

 

REQUIREMENTS:

  • S. with a minimum of 2 or B.S. with minimum of 4 years of relevant industry experience
  • Hands-on experience and knowledge of protein purification using HIC, IEX, SEC and affinity chromatography. Experience with GE AKTA FPLC is preferred
  • Excellent lab technique and the ability to work independently to analyze, interpret, and present data
  • Proficiency in biochemical and biophysical characterization methods (e.g. ELISA, DSC, SDS-PAGE, BLI, SPR) is desired
  • Ability to multi-task to support multiple projects across platform discovery organization
  • Ability to work in a highly collaborative, team-based environment
  • Excellent written and verbal communication skills

Preferred Experience and Skills:

  • Experience in developing, optimizing, and validating biophysical and biochemical assays for high-throughput screening applications
  • Experience with cell-based assays such as proliferation assays, apoptosis assays, and reporter gene assays.
  • Proficient with cell culture manipulation techniques (e.g., transfections, cell-based assays)
  • Prior experience with automation/liquid handling systems is a plus
